New Hope for Patients with Advanced Endometrial Carcinoma

A new study by Karyopharm Therapeutics offers promising results for women with advanced or recurrent TP53 wild-type endometrial carcinoma. The SIENDO study, presented at the ASCO Annual Meeting 2024, investigates the effect of Selinexor as maintenance therapy.

Study Results:

  • Progression-Free Survival (PFS): Patients who received Selinexor showed a median PFS of 28.4 months compared to 5.2 months in the placebo group.
  • Specific Subgroups: For TP53 wild-type/pMMR patients, the median PFS was 39.5 months (Selinexor) vs. 4.9 months (placebo).
  • Quality of Life: The Q-TWiST analysis showed a significant improvement in quality of life and duration in Selinexor-treated patients.

Safety: No new safety signals were observed. The most common side effects were mild to moderate nausea, vomiting, and diarrhea.

Dr. Reshma Rangwala of Karyopharm emphasizes that these results could position Selinexor as a potential new standard maintenance therapy. A further Phase 3 study will follow to confirm these promising results.
